OK let's review;
Wiring is correct and we have a nice spark on the sparkplug, you did make sure the engine run switch is on right?
New carb installed, do you have good gas flow to the carb? On your fuel tank does it have a manual shut off valve or what we call a vacuum petcock valve?
Muffler back on.
Adjusted the valves.
Try putting a little gas back in the spark plug hole and see what it'll do since the muffler is back on?

Then my only suggestion at this point is to pull the head and see if we have a stuck valve or one that is not seating or burnt.

I would take it apart first, one it may only need to have the valves lapped, very easy and inexpensive ( wife will love that) and two if you do need anything by taking it apart you can see what's wrong = no guessing.

Ok I'll do that today sand them down the valves but I do have to remember about the timing marks correct when the cylinder comes off because of the chain
