Have you checked to see that the carb is getting fuel? Also when is the last time you checked the valve clearance? The carb does have a drain that you can open to prove there is fuel. Whether you have a pump or a petcock they are both operated by vacuum.

List of possibilities;
If the manifold is broke it would have to be broken severly to keep it from at least trying to start, since you sprayed fluid in it and it'll run for a moment I'm doubting this.
Whether it's a fuel petcock or fuel pump both can go bad and if they have broken vacuum lines to them then no fuel so check all your lines.
Clogged fuel filter.
Extremely dirty air filter.
And as others mentioned needs the valves adjusted.

yes the new carb is getting fuel, the bowl was full. I loosen the drain screw and fuel is coming out. This is a brand new carb. I also have a clear fuel filter and gas is pumping through it. Everything is new lines, fuel filter. I just didn't check the air filter. It's a four stroke air cooled. JR you mention valve clearance how do I check that. I have no idea, but would like to find out. Also when i removed the new carb. you see a little fuel at the intake where the carb is installed, but the spark plug is dry.

As Alleyoop says, go .004 inch for the top (intake) valve and .005 inch for the bottom (exhaust) valve.
